Non-protected Synthesis of Oligonucleotides–N bond cleavage using 6-nitro-HOBt is more than 99% in the RNA synthesis without base protection. In this review, we summarize the .-selective phosphorylation in DNA and RNA synthesis without base protection and the synthesis of modified oligonucleotides having alkaline-labile functional groups using these new methods.

Site-Specific Modification of Nucleobases in Oligonucleotides, the site-specific modifications focusing on nucleobases within oligonucleotides are summarized. Moreover, as an experimental example of site-specific modification of an oligonucleotide, the preparation of .,.-disubstituted cytosine derivatives using reaction of 4-triazolylpyrimidin-2-one nucleobase with various secondary amines is demonstrated.
