Generation of Aneurogenic Larvae by Parabiosis of Salamander Embryosc larva and the aneurogenic limb (ANL) develops independently without innervation. Paradoxically, the limb in an ANL is capable of regeneration in a nerve-independent manner. Here, we describe a detailed method for the generation of ANL in the spotted salamander, ., for regeneration studies.

Derivation and Long-Term Culture of Cells from Newt Adult Limbs and Limb Blastemasion and maintenance of cell lines from adult post-metamorphic salamanders and in particular cells derived from normal and regenerating limbs. We also describe a protocol for nucleofecting newt cells that can be used both to investigate the gene function in short-term studies and to establish stable cell lines.

Isolation and Culture of Neurospheres from the Adult Newt Brainting and culturing of NSCs from the newt brain both during self-renewing and differentiating conditions. Newt NSC culture provides a useful tool for functional studies of NSC fate with the potential of resulting in novel regenerative strategies.
